IgG Purification Resin and Columns

Immunoglobulin G (IgG) Represents approximately 75% of serum antibodies in humans, IgG is the most common type of antibody found in blood circulation. IgG molecules are created and released by plasma B cells. Each IgG has two antigen binding sites. Human IgG1 and IgG3 are highly active in initiating Antibody Dependent Cell-Mediated Cytotoxicity (ADCC) and Complement Dependent Cytotoxicity (CDC). In contrast, IgG2 and IgG4 are much less capable in evoking such an immune response. As a result, IgG1 and IgG3 are the most suitable for therapeutic use against cancer cells.

Sheep IgGs against target proteins are often used as primary antibodies in various research applications. Thus, secondary anti-sheep IgGs conjugated to a detectable substrate are useful tools for the analysis of target proteins.

Sheep Serum Yield and Purity

Sheep IgG was purified from 750 microliters of sheep serum, 450 microliters of water and 300 microliters of sample diluent creating 1.5 ml at 50% serum for loading over LigaTrap® Sheep Purification resin.  It was compared to 750 microliters of sheep serum with 750 microliters of PBS loaded over Pierce Protein A Plus, and Thermo Protein G.  A five-minute residence time was used. Yields in histogram were determined by HPLC. All manufacturer recommended chromatographic protocols were followed.
